Belonging to the category of third-generation selective oral aromatase inhibitors, Arimidex or Anastrozole is an aromatase inhibitor used by some athletes and bodybuilders as part of their steroid cycles for reducing or preventing estrogenic side effects like bloating, oily skin, acne, and gynecomastia.

A single tablet of Arimidex contains 1 mg of Anastrozole, a non-steroidal aromatase inhibitor, and lactose, magnesium stearate, hydroxypropylmethylcellulose, polyethylene glycol as the inactive ingredient. The drug is chemically described as 1,3-Benzenediacetonitrile, a, a, a’, a’-tetramethyl-5-(1H-1,2,4-triazol-1-ylmethyl) and is freely soluble in methanol, acetone, ethanol, and tetrahydrofuran, and very soluble in acetonitrile. This potent and selective non-steroidal aromatase inhibitor has the molecular formula of C17H19N5.

One of the biggest advantages associated with this drug is that it prevents excess testosterone from aromatizing into estrogen while other drugs are meant for blocking, destroying, occupying existing estrogen receptors, or lowering sex hormone binding globulin that in turn inhibits estrogen formation. Arimidex also has the unique potential of increasing “free” testosterone [the amount of testosterone in the bloodstream after 2/3rd of non-free or bound testosterone is chemically bonded to a protein known as sex hormone-binding globulin (SHBG) and about 1/3rd to albumin, a protein]. Arimidex can significantly lower serum estradiol concentrations and has no detectable effect on the formation of adrenal corticosteroids or aldosterone. This drug also inhibits the conversion of androgens to estrogens in peripheral tissues (outside the central nervous system [CNS]), and a few CNS sites in different regions within the brain by binding reversibly to the aromatase enzyme through competitive inhibition.

Arimidex is used in daily doses of 1 mg (or 0.5-3.0 mg a day) with food and it may be used along with Propecia (Finasteride, a specific inhibitor of the 5a-reductase and known to reduce the serum concentration of DHT). This is because Finasteride can limit the transformation of Testosterone to Dihydrotestosterone besides reducing incidents of hair loss while Arimidex inhibits estrogen production. The dosages are usually taken two to six times a day at equal intervals as Anastrozole has a short active life. The ideal duration of Arimidex use is 4-6 weeks though most users prefer using Arimidex in post cycle therapy for seven to 14 days.
